Tecnology 5 - A Kit for Parentage Verification in Indian Ruminant Livestock

1.Current IP Status: Complete application filed on 4th February 2013.

2.Technology introduction and description: Parentage analysis of livestock animals is becoming increasingly important in animal husbandry to improve the accuracy of selection and to select sires to increase production and reproduction traits. We have developed a single parentage verification kit for all the ruminant livestock species which include cattle (B. indicus, B. taurus and B. indicus x B. taurus crosses), sheep (Ovis aries), goat (Capra hircus), yak (Bos grunniens), mithun (Bos frontalis) and buffalo (Bubalus bubalis). Till date no kit has been made to prepare a single kit meant for all the six livestock species. We selected a set of markers which amplified in six species of ruminant livestock which were cattle, sheep, goat, buffalo, yak and mithun. Eleven microsatellites were finally selected that could be simultaneously amplified in all the six ruminant species of livestock. The utility of the microsatellite markers were tested on populations of all the six livestock species. The probability of exclusion based on the set of 11 microsatellites was estimated in all the six livestock species.

3.Benefits, usefulness, problem solving, business opportunities from the technology to the Client(s):

  • This is the first kit based on DNA markers for amplification of 11 markers in six ruminant livestock species.
  • The kit utilises a single multiplex PCR reaction-having 11 genetic markers for the generation of genotypes of the individual in all the six ruminant livestock species.
  • The kit has been developed using a number of populations/breeds from different parts of the country. For common species like sheep, goat, cattle and buffalo large data set was generated to check the efficacy of the kit. For yak and mithun, the data set was generated from the geographical regions of India where these species exist. It has performed well in amplification of DNA from vast and varied geographical locations. All markers are polymorphic and exhibit heterozygosity.
  • The kit can be used for the parentage verification, providing individual identification and testing correctness of the pedigree information in all the six ruminant livestock species.
